Gene Therapy has emerged as a promising future to disease treatment and personalised therapies. Gene Therapy is the introduction or modification of genes that modifies gene function to prevent or cure diseases.

AAV based Gene Therapy

In vivo gene therapy involves direct delivery of a therapeutic gene into the target cell in a patient's body. This can be carried out by both viral and non viral vectors, though the most popular emergence in vectors has been the Adeno-Associated Virus vector (AAV). AAV binds to a cellular receptor, enters the cell, migrates to the nucleus, and delivers a single-stranded DNA genome to establish a latent state for long-term gene expression in the absence of coinfection with a helper virus.

AAV vectors are commonly used in optogenetics experiments. These viruses are preferred over lentiviruses because they remain primarily episomal, while lentiviruses integrate into the genome. This is important because local chromatin structure at the site of genome integration can affect the expression of transgenes, as well as the expression of neighboring genes. Eleven serotypes of AAV have thus far been identified, with the best characterized and most commonly used being AAV5. These serotypes differ in their tropism, or the types of cells they infect, making AAV a very useful system for preferentially transducing specific cell types.

Krishgen has a range of titration AAV ELISA kits for a rapid and reliable way of quantifying AAV vectors based on a monoclonal antibody sandwich technique. And a range of Anti-AAV ELISA kits for qualitative determination of antibodies to AAV5 in serum and plasma.

Dendritic Cells

DCs represent unique antigen-producing cells capable of sensitizing T cells to antigens. They play a central role in the regulation of innate and adaptive immunity. DC therapy represents a new and promising immunotherapeutic approach for treatment of different types of cancer.
